thrombin-induced modifications of human platelet cytoskeleton

چکیده
1. Thrombin treated with phenylmethanesulphonyl fluoride, like active enzyme, promotes modifications to human platelet cytoskeleton. 2. The removal of active thrombin by hirudin partially reverses this process. 3. Chymotrypsin-treated platelets do not modify their cytoskeleton after thrombin stimulus, but are still able to increase their adhesiveness to collagen. It is concluded that thrombin influences the cytoskeleton and adhesion by non-enzymic mechanisms which may be mediated by different modulators.

Escitalopram Impairs Thrombin-Induced Platelet Response, Cytoskeletal Assembly and Activation of Associated Signalling Pathways.
Background Serotonin reuptake inhibitors (SSRIs) may impair platelet function. Thrombin is a strong platelet agonist causing irreversible aggregation, release of granules' contents, cytoskeletal rearrangement and activation of signalling pathways. We investigated the effects of the SSRI escitalopram (SCIT) on thrombin-induced platelet response. Methods Isolated platelets were exposed to SCIT a...
متن کاملAssociation of fibrin with the platelet cytoskeleton.
We have previously postulated that surface membrane proteins become specifically associated with the internal platelet cytoskeleton upon platelet activation (Tuszynski, G.P., Walsh, P.N., Piperno, J., and Koshy, A. (1982) J. Biol. Chem. 257, 4557-4563). Four lines of evidence are in support of this general hypothesis since we now show that platelet surface receptors for fibrin become specifical...
متن کاملThrombin-induced GPIb-IX centralization on the platelet surface requires actin assembly and myosin II activation.
In resting platelets, the GPIb-IX complex, the receptor for the von Willebrand factor (vWF), is linked to underlying actin filaments by actin-binding protein (ABP-280). Thrombin stimulation of human platelets leads to a decrease in the surface expression of the GPIb-IX complex, which is redistributed from the platelet surface into the open canalicular system (OCS). Because the centralization of...
متن کاملIQGAP2 functions as a GTP-dependent effector protein in thrombin-induced platelet cytoskeletal reorganization.
Human blood platelets are anucleate cells whose response to extracellular stimuli results in actin cytoskeleton rearrangements, thereby providing the critical initial step in the regulation of hemostasis. The serine protease alpha-thrombin, known to activate platelets by cleavage of a family of protease-activated receptors (PARs), is the most potent physiologic activator of human platelets, tho...

Abstract Background Autologus platelet gel is easy to prepare and is relatively low cost. The aim of this study was to prepare and evaluate in vitro efficacy of autologus platelet gel. Materials and Methods In this experimental study, platelet concentrate and platelet poor plasma were prepared with aphaeresis method. Thrombin was prepared by mixing the plasma with calcium. Thrombin activity...
